[Adverse effects of the vaccines Tetracoq, IPAD/DTCP and DTCP. A French study of regional drug monitoring centers]

On request of the French Drug Agency, the Regional Pharmacovigilance Center (RPVC) of Tours has been in charge of the analysis of adverse events (AEs) associated with tetravalent vaccines IPAD/DTCP, DTCP and Tetracoq, and reported to the RPVC or to the pharmaceutical companies that produce them. METHODS: All AEs spontaneously reported during use of one of these vaccines to one of the French Pharmacovigilance Centers or to the responsible firms between January 1, 1986 and December 31, 1990 were take into account. An AE was noted as "serious" in accordance with the European criteria. The incidence of adverse effects was estimated by evaluating the ratio of adverse effects and the number of sales of the vaccine for the same period. RESULTS: From 1986 to 1990, 631 AEs (with 19 duplicate cases) associated with tetravalent vaccines in 606 children (75% < 1 year) were reported. The most frequent AEs were: local AEs at the site of injection (43%), neurologic disorders (12%), hyperthermia (10%) and allergic reactions (10%). Serious AEs represented 25% of all AEs and were similar to those usually described with these vaccines, particularly persistent crying (23), febrile seizures (12), apyretic seizures (14), uneasiness (28) and, rarely, shock (3). CONCLUSION: Incidences of AEs reported with pentavalent vaccines are very low, probably underestimated because of the under-notification by prescribers of AEs of vaccines licensed some time ago. It will be interesting to compare these data with AEs of penta- and hexavalent vaccines since they have replaced tetravalent vaccines.

Childhood bullous pemphigoid developed after the first vaccination.

Bullous pemphigoid (BP) typically affects the elderly. There are at least 40 reported cases of BP in childhood, 10 reported cases at 1 year of age or younger. The antigen of this autoimmune disease is localized to the hemidesmosome. Neoplasia, recurrent trauma, some systemic diseases, and psoriasis were previously reported as possible triggering factors of bullous pemphigoid in some cases. In the last 5 years, 10 adult and 2 infantile BP cases with a close relation of vaccination have been reported. Anti-influenza vaccine, tetanus toxoid booster, and tetracoq vaccine were the possible causes of these cases. We report herein a 3.5-month-old BP case in whom the lesions developed 24 hours after the first tetracoq vaccine. We suggest that vaccination may be the triggering factor of BP of any age by stimulating the immune system with an unexplained mechanism.

[Post-vaccination granuloma due to aluminium hydroxide]

Post-vaccination granulomas a well-known reaction due to aluminium adsorbed vaccines. We report three cases of children who developed subcutaneous nodules at the site of a previous injection of Tetracoq*vaccine (tetanus, diphtheria, Bordetella pertussis, poliovirus). Histologically, the lesions were characterized by a necrotizing granulomatous reaction with eosinophilic crystalline material. This material stained positively with the solochrome cyanine stain and was pink-purple.This aluminium stain enabled diagnosis of post-immunization injection-site reaction due to aluminium.

следующая - продукт российских ученых. Может быть Вам она понравится, но меня немножко покоробила. Сомневаюсь в честности исследователей. А еще меня заинтересовал вопрос - первая статья по тетракоку - начало 90хх. Почему-то во Франции в то время такое исследование выполнено не было. Правда, допускаю, что это из соображений облегчения лицензирования? Может такое быть?

[The vaccination of children with severe somatic pathology]

1,696 children were vaccinated; of these, 1,487 children had different kinds of somatic pathology, including 1,181 children with CNS lesions, 29 children with malignant tumors, 45 children with congenital defects, 82 children with allergic diseases, etc. The group of relatively healthy vaccinees consisted of 209 children. The following vaccines were used for immunization: Tetracoq 05, D.T.Vax, Rudivax, Imovax Polio, Vaxigrip (Pasteur Merieux Connaught, France); HBVax, MMRII (Merck Sharp & Dohme, USA); as well as vaccines against hepatitis B produced by Smith Kline Beecham (UK) and Combiotech (Russia). In no case severe vaccine-associated complications were observed. The frequency and manifestation of reactions in children with somatic pathology did nor essentially differ from those in relatively healthy children. The increase of the number of vaccine components did not lead to the increase of the number of side effects of the severity of their manifestation. These investigations demonstrated the safety of vaccination for children with somatic pathology.

A study of the tolerance and potency of a quadruple vaccine adsorbed on calcium phosphate was carried out in Senegal. After a follow up of four months the local tolerance was better when the vaccine was inoculated by intramuscular injection. A high level of passively acquired antibodies was found for the three subtypes of polioviruses and seemed to delay the immunological reaction. 90%, 96% and 92% of the total group of children involved in this study had a positive reaction.
